Dimerization mutants of alcohol dehydrogenase of maize.

چکیده

Two ethyl methanesulfonate-induced mutants of the alcohol-dehydrogenase gene in maize have been analyzed. The enzyme subunits produced by the mutants seem to be structurally altered in such a way that the subunit specified by the null allele Adh(1) (0(5657)) does not dimerize with "wild-type" subunits, and that specified by Adh(1) (S(296)) does so only to a limited extent.

The molecular basis for allelic complementation of alcohol dehydrogenase mutants of maize.

Studies on interallelic complementation with two temperature-sensitive alcohol dehydrogenase mutants in maize are described. The data suggest that the phenomenon results from instability or abnormal maturation of the mutant homodimer rather than correction of configurational defects in mutant heterodimers.

A pollen selection system for alcohol-dehydrogenase-negative mutants in plants.

Exposure of freshly shed maize pollen to allyl alcohol vapors allows selective fertilization by mutant alcohol-dehydrogenase-negative grains. Wild-type pollen grains are killed by the enzymatic conversion of allyl alcohol to the highly toxic acrylaldehyde.

Brown midrib mutations and their importance to the utilization of maize, sorghum, and pearl millet lignocellulosic tissues

Brown midrib mutants have been isolated in maize (Zea mays), sorghum (Sorghum bicolor) and pearl millet (Pennisetum glaucum) arising by either spontaneous or chemical mutagenesis. The characteristic brown coloration of the leaf mid veins is associated with reduced lignin content and altered lignin composition, traits useful to improve forage digestibility for livestock. Brown midrib phenotype i...

Differential expression of alternative oxidase genes in maize mitochondrial mutants.

We have examined the expression of three alternative oxidase (aox) genes in two types of maize mitochondrial mutants. Nonchromosomal stripe (NCS) mutants carry mitochondrial DNA deletions that affect subunits of respiratory complexes and show constitutively defective growth. Cytoplasmic male-sterile (CMS) mutants have mitochondrial DNA rearrangements, but they are impaired for mitochondrial fun...

Adh locus in maize for detection of mutagens in the environment.

The advantages of the maize Adh system for low level mutagenesis studies are discussed. They include high mutation rate and the availability of both a strong selection system for null mutants and a sensitive method for detection of variants. The null selection involves treatment of pollen with allyl alcohol which is converted by the Adh enzyme to the toxic acrylaldehyde. Adh variants can be det...
